STEP BY STEP GUIDE ON HOW TO PREPARE POTATO DEXTROSE AGAR

 

Prepare materials

  1. Potato Starch – 20g
  2. Agar Powder – 15g
  3. Sugar – 15g
  4. Yeast (Baking Powder) – 1g
  5. ‘Non-Chlorine’ water – 1.5l
  6. Plastic Container (SKP-T5 PP5) – 12 cups
  7. Ladle
  8. Sieve
  9. Tin Vessel
  10. Stirrer
  11. Weighing Scale
  12.  Pot

 

How to

  1. Wash & Clean utensils and containers thoroughly
  2. Label the containers by Date & Time (Optional)
  3. Start stove and put 1.5l ‘Non-Chlorine’ water to boil
  4. Put all ingredients into the tin vessel fill it with some water
  5. Stir & mix all ingredients thoroughly until its dissolved
  6. Pour mixed ingredients over a sieve into a clean vessel
  7. Gently pour mixed ingredients over the pot of boiling water
  8. Continue to stir the pot of ingredient mixture for 5-10 minutes
  9. Lower the fire and continue to stir for 3-5 minutes
  10. Turn off the stove
  11. Pour 15-20ml of the mixture into the 12 containers
  12.  Cover it with a lid

 

Autoclave Process

  1. Fill the pressure cooker or steamer with the RIGHT amount of water
  2. Please watch the video for the amount of water required for a pressure cooker
  3. Take note of the TIME you start autoclaving
  4. Autoclaving can stop between 45 mins – 60 mins
  5. Turn off the stove
  6. DO NOT OPEN THE LID after you turn off the stove
  7. Remove the autoclave lid after 1 hour later
  8. Remove the containers and keep them to rest
  9. Occasionally, the container can be ‘DEFORMED’ but still can be used
  10. Leave the container to rest for 3-5 days before tissue culture
